I am mounting a small Lister 3.5HP @1500 rpm onto a concrete block, I have seen stationary engines with a hardwood block between engine base and concrete block and some without.

I can only guess it provides a little dampening and or as is mostly the case the flywheel projects below the base and the hardwood block represents a spacer.

Any opinions on this point?

I've heard if you mount a diesel, especially a single cylnder, on concrete, the vibration can travel a long way. If you house/shed is on/near the concrete pad, then the whole building becomes a sounding board.

Wood blocks would help, but rubber would be better. Got any old tires you can cut up? Or some old vehicle engine mounts.

Hi,
In addition to what Glen has suggested There is a problem with condensation causing rust or corrosion, the wood barrier is an insulator to prevent electrolysis with the earth. Edit; Best design is build a small Skid and and mount on rubber pads.
